Abstract 

Nanoparticles have been extensively analyzed for its potential use in the biomedical application. The present study was aimed to investigate the bio-activity of ZnS nanoparticles in normal MDCK cells and cervical carcinoma HeLa cells. MDCK and HeLa cell viability was assessed by the Sulphorhodamine-B (SRB) assay following ZnS nanoparticles exposure. ZnS nanoparticles showed significant reduction in HeLa cell viability, while it has a marginal effect on normal MDCK cells. Morphologically HeLa cells were changed significantly, whereas in MDCK cells, no remarkable changes were observed following nanoparticles exposure. The mRNA expression of apoptotic gene p53, and caspase 3 was up-regulated in the HeLa cells, whereas in MDCK cells, a marginal increase was found. Mitochondria-derived reactive oxygen species (ROS) was significantly increased in HeLa cells, and lesser degree in MDCK cells. Malondialdehyde (MDA) was measured as an index of lipid peroxidation by TBARS method. MDA content was significantly increased in the cancer cells, while it was not significantly altered in MDCK cells following ZnS nanoparticle exposure. Taking all these data together, it may be concluded that the ZnS nanoparticle may have a strong bio-potential in the cervical carcinoma HeLa cells than normal MDCK cells.
